Anti-Methicillin Resistant Staphylococcus aureus (MRSA) of Methanol Extract of Mangrove Plants Leaf: Preliminary Report

ABSTRACT

Increasing multidrug resistant (MTDR) in bacteria, in particular Staphylococcus aureus has been becoming worst. Finding new antibiotic source for MTDR is compulsion for scientist and clinicians due to the limitation of the therapeutic option. This research was conducted to investigate the antibacterial activity of methanol extracts of Sonneratia caseolaris, Acanthus ilicifolius, Rhizophora mucronata and Excoecaria agallocha against methicillin resistant, clinical isolates of Staphylococcus aureus using disk diffusion method. The minimum inhibitory concentration (MIC) values of all methanolic mangrove leaves ranged from 6 to 20 mg/mL. The mangrove leaf of Excoecaria agallocha possessed higher anti MRSA potency than the other mangrove plants. It is clear that mangrove leaves especially, E. agallocha leaf has anti MRSA compound. Further purification of active compound from the potent mangrove will be useful for the control of drug resistant strains of S. aureus.

Phytochemical screening and Antibacterial activity of methanol extract of mangrove plant (Rhyzophora mucronata) from Porong River Estuary

Abstract

The aims of this study were to obtain phytochemical content and to examine antibacterial activity potency of methanol extract of Rhyzophora mucronata. The phytochemical screening was assayed by qualitative analysis and the antibacterial assay determined by disk diffusion method. The results of phytochemical analysis showed that all methanol extract of root, bark, leaf, fruit and flower of R. mucronata contained alkaloid, tannin, and flavonoid, in addition fruit contained steroid., respectively. The antibacterial activity in gram positive bacteria (Staphylococcus aureus) and gram negative bacteria (Escherichia coli) was showed by almost all extracts of root, bark, wood, flower and leaf of R. mucronata.

PERBANDINGAN AKTIVITAS ANTIBAKTERI Penicillium notatum ATCC 28089 DENGAN Penicillium sp. R1M YANG DIISOLASI DARI MANGROVE Sonneratia caseolaris

Abstract

Mangrove Sonneratia caseolaris is a potential host for  endhopytic fungi that commonly produce strong dan unique antibiotic. The objective of the research was to compare the antibacterial activity of Penicillium notatum ATCC 28089 and endophytic fungus Penicillium sp. R1M isolated from Mangrove Sonneratia caseolaris. First step of the research was observation the growth curve of each isolate. Second step was isolation of their  metabolites to be used in the antibacterial activity assay against Staphylococcus aureus ATCC 9144 and Escherichia coli ATCC 8739. The results showed that both isolates indicated different growth curves. Penicillium sp. R1M experienced shorter adaptation phase, than P. notatum  ATCC 28089.  The secondary metabolites of  P. notatum ATCC 28089 was isolated on day 8, while those of Penicillium sp. R1M isolated on day 6. The result of antibacterial assay  confirmed  that the  extracts of mycelia Penicillium sp. R1M show inhibition diameter in the growth of S. aureus and E. coli  with 11.5 ± 0.6 mm and 10.6 ± 0.4, mm, respectively, while the medium extracts of  P. notatum ATCC 28089 were 9.3 ± 0.9 mm and 7.4 ± 0.1 mm, respectively. The  inhibitory zone diameter of  medium extracts. Penicillium sp. R1M against S. aureus and  E. coli were correspondingly 7.5 ± 0.1 mm and 7.1 ± 0.8 mm, while  the medium  extracts of P. notatum ATCC 28089 were 9.3 ± 0.6 mm and 7.4 ± 0.4 mm, respectively. Mycelial extracts of  Penicillium sp. R1M showed better antibacterial activity than those of  P. notatum ATCC 28089. On the other hand the medium extracts showed the opposite results.

Aktivitas Antimalaria (Plasmodium falciparum 3D7) dari Spon Laut Acanthella sp

Abstract

Penelitian ini bertujuan untuk mendapatkan kandidat bioaktif anti multiresisten malaria dari bioaktif spon laut Acanthella sp.. Tahap penelitian diawali dengan mengisolasi senyawa dari Acanthella sp.. Selanjutnya dilakukan uji antimalaria secara invitro terhadap Plasmodium falciparum 3D7. Pengujian terhadap P. falciparum strain 3D7 menggunakan dosis konsentrasi bertingkat dengan kontrol positif digunakan chloroquin. Perhitungan penghambatan skizon dilakukan dengan pewarnaan darah dan selanjutnya dihitung jumlah eritrosit yang terinfeksi per 1000 eritrosit. IC50 dihitung menggunakan analisis probit. Hasil penelitian menunjukkan bahwa keempat fraksi aktif menunjukkan penghambatan pertumbuhan P. Falciparum dengan nilai IC50 yang berbeda-beda. Nilai terbaik dihasilkan oleh fraksi B dengan IC50 sebesar 0,013 ppm.

Endophytic Fungi Isolated from Mangrove (Rhyzopora mucronata) and Its Antibacterial Activity on Staphylococcus aureus and Escherichia coli

Abstract: Disc diffusion method was used to investigate the antibacterial activity of methanol extract of five endophytic fungi isolated from mangrove (Rhizopora mucronata) on Staphylococcus aureus ATCC 9144 andEscherichia coli ATCC 8739. Fungi were isolated from leaf, stem and root of R. mucronata. Potato dextrose agar and potato dextrose broth were used for fungi purification and fermentation. Fungal metabolites were extracted with methanol (1:1 v/v) for 24 h before vacuum evaporated to yield the crude extracts. Antimicrobial activity test was done according to the Kirby-Bauer test. The inhibition zone around each disc was considered to measure antibacterial activity of endophytic fungi. Only three endophytic fungi showed inhibition activity on S. aureus ATCC 9144 and two endophytic fungi showed inhibition on E. coli ATCC 8739. The results suggested that the extracts have a high potential for enteric diseases treatment caused by foodborne pathogen.

PENAPISAN ANTIBAKTERI DAN FITOKIMIA EKSTRAK METANOL TANAMAN MANGROVE (Avicennia mucronata) DARI MUARA SUNGAI PORONG

Abstrak

Tujuan penelitian ini adalah untuk mendapatkan kemampuan aktivitas antibakteri dan kandungan fitokimia ekstrak metanol Avicennia mucronata. Uji aktivitas antibakteri dilakukan dengan metode difusi agar tuang dan uji fitokimia dengan analisis kualitatif. Hasil penelitian menunjukkan bahwa ekstrak metanol akar, batang, daging buah, kulit kayu, bunga dan daun  Avicennia mucronata dapat menghambat pertumbuhan bakteri gram positif (Staphylococcusaureus) dan gram negatif (Escherichiacoli). Hasil fitokimia menunjukkan semua ekstrak dari akar, batang, kulit kayu, bunga dan daun  Avicennia mucronata mengandung senyawa alkaloid, tanin dan flavonoid, serta buah juga mengandung steroid.

CYTOTOXIC, ANTIOXIDANT AND ANTIBACTERIAL ACTIVITY OF METHANOL EXTRACT OF Xylocarpus moluccensis FRUIT HUSK

Abstract

The aims of this study were to examine toxicity, antioxidant and antibacterial activity of methanol extract of fruit husk (Xylocarpus moluccensis). Toxicity assay was undertaken by brine shrimp lethal toxicity test, antioxidant activity was done by Diphenyl picryl hydrazil (DPPH) scavenging, and inhibition growth to Staphylococcus aureus and Escherichia coli used as antimicrobial test. The results showed that methanol extract of fruit husk was not toxic, antiradical efficiency was 0.000154, and broad spectrum antibacterial with inhibition zone to Staphylococcus aureus and Escherichia coli was 12.7±1.2  and 11.9±0.9 mm, respectively in 630000 ppm concentration.

ISOLASI BAKTERI PENGHAMBAT QUORUM SENSING DARI PERAIRAN TAMBAK DENGAN MENGGUNAKAN BIOSENSOR Chromobacterium violaceum CV026

Abstrak

Quorum sensing bakteri mempunyai beragam implikasi terhadap tingkah laku bakteri. Quorum sensing diketahui berhubungan dengan proses patogenisitas, virulensi serta pembentukan biofilm dan perubahan DNA. Proses quorum sensing ini dapat dihambat dengan menggunkan bakteri yang mampu menghalangi proses quorum sensing yaitu proses quorum quenching. Bakteri quorum sensing inhibitor (QSI) didapatkan dengan mengisolasi sedimen dan air dari tambak udang tradisional, semi intensif dan intensif. Isolasi bakteri dilakukan dengan menumbuhkan sampel kedalam media NA. setiap koloni yang terbentuk kemudian dilakukan kultur murni untuk pengujian penghambatan quorum sensing menggunakan biosensor Chromobacterium violaceum CV026. Hasil dari pengujian penghambatan quorum sensing didapatkan dua spesies bakteri yang mempunyai kemampuan melakukan penghambatan terhadap quorum sensing. Isolat bakteri yang mempunyai penghambatan terbesar dilanjutkan dengan analisis spesies menggunakan MicrobactTM dan dikonfirmasi melalui analisis 16S rDNA. Hasil analisis menunjukkan bahwa spesies bakteri yang mempunyai kemampuan penghambatan quorum sensing terbesar adalah Vibrio parahaemolyticus strain LCL28209.
